报告国内首次建立两株人胰腺癌裸小鼠胰腺内原位移植模型。方法系将人胰腺癌手术标本直接移植于BALB/C-nu/nu裸小鼠胰腺内,肿瘤异体移植和连续传代获得成功。研究了其生物学特性。结果表明:PTNMP-1已传至9代,PTNMP-2已传至6代。肿瘤移植成功率为95%~100%。两株移植瘤均能产生大量的癌胚抗原CEA,染色体分析证实为人类肿瘤细胞核型。在荷瘤裸小鼠体内出现淋巴道、血道转移,光镜、电镜检查结果证明原位移植瘤的形态学与原手术标本完全一致。此模型可用于胰腺癌的侵袭、转移及实验治疗。
Reported that the first domestic establishment of two pancreatic cancer nude mice in pancreas orthotopic transplantation model. Methods The human pancreatic cancer surgical specimens were directly transplanted into the pancreas of BALB/C-nu/nu-nude mice. Allogeneic tumor transplantation and serial passages were successfully performed. Its biological characteristics were studied. The results showed that: PTNMP-1 has been transmitted to the 9th generation, and PTNMP-2 has been transmitted to the 6th generation. The success rate of tumor transplantation was 95% to 100%. Both transplanted tumors can produce a large number of CEA antigens. Chromosomal analysis confirmed the karyotype of human tumor cells. The lymphatic and hematogenous metastasis were observed in the tumor-bearing nude mice. Light microscopy and electron microscopy showed that the morphology of the orthotopic transplanted tumor was completely consistent with the original surgical specimen. This model can be used for invasion, metastasis and experimental treatment of pancreatic cancer.
