目的:探讨脂联素(ADPN)对体外高糖培养的人肾小管上皮细胞转化生长因子-β1(TGF-β1)表达的影响,以揭示ADPN对高糖环境中的肾小管上皮细胞的可能保护作用机制。方法:人肾小管上皮细胞分别培养于不同的培养基中,在高糖环境中加入脂联素,分别培育48 h后采用酶联免疫吸附法(ELISA)测定细胞上清液中TGF-β1的含量。结果:TGF-β1在高糖组表达较高,加入中等浓度(ADPN浓度为10.0,15.0,20.0μg/mL)TGF-β1的表达减少,和高糖组相比,两者的差异有统计学意义。结论:中等浓度ADPN可以抑制TGF-β1在人肾小管上皮细胞的表达。
Objective: To investigate the effect of adiponectin (ADPN) on the expression of transforming growth factor-β1 (TGF-β1) in cultured human renal tubular epithelial cells cultured in high glucose to reveal the possible protective effect of ADPN on renal tubular epithelial cells Mechanism. Methods: Human renal tubular epithelial cells were cultured in different media. Adiponectin was added in high glucose environment. After cultured for 48 h, the expression of TGF-β1 in the supernatant of the cells was determined by enzyme-linked immunosorbent assay (ELISA) content. Results: The expression of TGF-β1 in high glucose group was higher than that in high glucose group, and the expression of TGF-β1 in medium concentration group (10.0, 15.0 and 20.0 μg / mL ADPN) was decreased significance. CONCLUSION: Moderate ADPN can inhibit the expression of TGF-β1 in human renal tubular epithelial cells.
