目的分析约氏疟原虫感染后小鼠红细胞膜蛋白及受染红细胞与内皮细胞黏附能力的变化,为进一步探索脑型疟的发生机制提供线索。方法提取约氏疟原虫感染的BALB/c小鼠红细胞膜蛋白,进行SDS-PAGE电泳,检测红细胞膜蛋白的变化。用内皮细胞株与小鼠红细胞共同培养,观察受染红细胞与内皮细胞黏附能力的改变。结果 SDS-PAGE显示约氏疟原虫感染后的小鼠红细胞膜蛋白中含有分子质量约137ku组分,53/54ku固有膜蛋白减少。与未感染对照组比较,受染红细胞对内皮细胞的黏附能力增强约3倍,差异有统计学意义(P<0.05)。结论疟原虫感染可改变宿主红细胞膜蛋白组成,增强受染红细胞与内皮细胞的黏附力。这些变化可能与脑型疟的发生有关。
Objective To analyze the changes of erythrocyte membrane protein and adhesion of erythrocytes and endothelial cells after Plasmodium yoelii infection in mice and provide clues for further exploration of the pathogenesis of cerebral malaria. Methods Erythrocyte membrane proteins of BALB / c mice infected with Plasmodium yoelii were extracted and analyzed by SDS-PAGE electrophoresis to detect the changes of membrane proteins of erythrocytes. Endothelial cells were co-cultured with mouse erythrocytes to observe the changes of adhesion ability of infected erythrocytes and endothelial cells. Results SDS-PAGE showed that the erythrocyte membrane protein of Plasmodium yoelii infected mice contained 137 ku molecular weight and 53/54 ku lamina propria protein decreased. Compared with non-infected control group, the adhesion ability of infected erythrocytes to endothelial cells increased about 3-fold, the difference was statistically significant (P <0.05). Conclusion Plasmodium infection can change the protein composition of the host erythrocyte membrane and enhance the adhesion of the infected erythrocytes to the endothelial cells. These changes may be related to the occurrence of cerebral malaria.
