目的检测TSP-1在早期糖尿病大鼠视网膜组织中的表达并探讨其作用。方法采用链脲佐菌素(STZ)诱导大鼠糖尿病模型,成模后随机分成两组,糖尿病组和多贝斯组,其中,多贝斯治疗组在出现糖尿病表现后按100mg/kg/d,糖尿病组和正常对照组只在同期灌胃等量生理盐水。注射后1、3个月采用免疫组织化学方法检测视网膜中TSP-1阳性反应及其分布。结果各组糖尿病大鼠视网膜TSP-1蛋白质阳性着色的积分光密度值均较正常组降低(P<0.05)。结论 TSP-1蛋白质在糖尿病大鼠视网膜中的表达明显降低并与病程有关,提示TSP-1表达不足可能是糖尿病视网膜病变的原因之一。
Objective To detect the expression of TSP-1 in retina of early diabetic rats and to explore its role. Methods Diabetic rats were induced by streptozotocin (STZ). The rats were randomly divided into two groups: diabetic group and domperidone group. Diabetes group and diabetic group Group and normal control group only the same period of gavage with saline. At 1 and 3 months after injection, immunohistochemistry was used to detect the positive TSP-1 in the retina and its distribution. Results The integral optical density of TSP-1 protein positive staining in retina of diabetic rats was lower than that of normal group (P <0.05). Conclusions The expression of TSP-1 protein in the retina of diabetic rats was significantly decreased and correlated with the course of the disease. It suggests that the insufficient expression of TSP-1 may be one of the causes of diabetic retinopathy.
