目的探讨SCUBE3在骨肉瘤细胞和组织中的表达及其与患者预后的关系。方法利用Western blotting技术检测5种骨肉瘤细胞株SCUBE3的表达,利用化学合成siRNA转染骨肉瘤细胞,靶向抑制SCUBE3表达。CCK8细胞增殖实验观察SCUBE3对U2OS细胞增殖的影响,流式细胞仪检测细胞周期改变。免疫组化检测SCUBE3在骨肉瘤组织中的表达,并运用Kaplan-Meier法进行生存分析。结果 SCUBE3在骨肉瘤细胞株中的表达均高于正常成骨细胞。干扰SCUBE3表达能抑制U2OS的生长(P<0.05),同时细胞周期阻滞在G1期。Kaplan-Meier生存分析显示,SCUBE3高表达的患者预后比低表达组差(P=0.036)。结论 SCUBE3在骨肉瘤中高表达,它能促进骨肉瘤细胞的增殖并与患者不良预后相关,提示其可能成为潜在的治疗靶点。
Objective To investigate the expression of SCUBE3 in osteosarcoma cells and tissues and its relationship with the prognosis of patients. Methods Western blotting was used to detect the expression of SCUBE3 in 5 osteosarcoma cell lines. The siRNA was transfected into osteosarcoma cells to inhibit the expression of SCUBE3. The effect of SCUBE3 on the proliferation of U2OS cells was observed by CCK8 cell proliferation assay. The cell cycle was detected by flow cytometry. The expression of SCUBE3 in osteosarcoma was detected by immunohistochemistry and the survival analysis was performed by Kaplan-Meier method. Results The expression of SCUBE3 in osteosarcoma cell lines was higher than that in normal osteoblasts. Interference with SCUBE3 expression inhibited the growth of U2OS (P <0.05), and the cell cycle arrest was in G1 phase. Kaplan-Meier survival analysis showed that patients with high SCUBE3 expression had a worse prognosis than those with low expression (P = 0.036). Conclusions SCUBE3 is overexpressed in osteosarcoma, which can promote the proliferation of osteosarcoma cells and correlate with poor prognosis of patients, suggesting that SCUBE3 may be a potential therapeutic target.
