目的:研究南澳岛海域亨氏马尾藻多糖的提取工艺,并对其进行纯化和鉴定。方法:用热水提取多糖,乙醇沉淀,并用正交实验确定提取的最佳条件,Sevage法去蛋白,用CTAB结合不同浓度盐离子溶液对多糖进行分级。SephadexG200柱层析及聚丙烯酰胺凝胶电泳进行纯度鉴定。硅胶薄层层析分析多糖中单糖组分。结果:最佳提取条件:100℃,pH7,提取4h,得率可达22.7%,分级沉淀后得四组分F1、F2、F3、P,纯度鉴定证明其中F1、F2、P为均一级分,薄层层析发现其中含有岩藻糖。结论:亨氏马尾藻多糖的分离、纯化和鉴定能够为该多糖的进一步开发利用提供一定的理论依据。
Objective: To study the extraction process of Sargassum hesmeensis polysaccharides from Nan’ao Island and to purify and identify them. Methods: The polysaccharides were extracted with hot water and precipitated with ethanol. The optimum extraction conditions were determined by orthogonal experiment. The protein was degraded by Sevage method. The polysaccharides were fractionated by CTAB combined with different concentrations of salt ion solution. Sephadex G200 column chromatography and polyacrylamide gel electrophoresis for purity identification. Thin-layer chromatography on silica gel was used to analyze the monosaccharide components in polysaccharides. Results: The best extraction conditions: 100°C, pH7, extraction for 4h, yield up to 22.7%, four fractions F1, F2, F3, P obtained after fractionation, purity identification proves that F1, F2, P are homogeneous. Fractions were found on thin layer chromatography containing fucose. Conclusion: The separation, purification and identification of Sargassum hepatica polysaccharides can provide a theoretical basis for the further development and utilization of polysaccharides.
