采用普洛麦格公司 Wizard TM染色体 DNA纯化试剂盒提取牙龈卟啉菌、伴放线放线杆菌、中间型普氏菌、产黑色素普氏菌、躯体普氏菌五种牙周可疑致病菌的 DNA,获得浓度为 687～ 14 5 6μg/ml的细菌染色体 DNA,OD2 6 0 /OD2 80 值介于 1.75～ 1.85之间。电泳结果显示 ,DNA分子量均大于 2 30 0 0个碱基对 ( 2 3kb)。表明该方法能在短时间内获得高纯度及比较完整的牙周细菌染色体 DNA,与传统的酚 /氯仿抽提方法相比 ,是一种高效而实用的新方法。
