研究神经元特异性烯醇化酶(NSE)、胶质纤维酸性蛋白(GFAP)及Notch1受体蛋白在中药大黄三七散改善急性脑出血(ICH)大鼠神经功能缺损中的表达及其意义。以肝素胶原酶Ⅳ法制作大鼠脑出血模型,考察了施药前后ICH大鼠神经功能评分、脑系数及脑含水量、NSE、GFAP及Notch1在mRNA及蛋白水平的表达量及相关性。结果表明,大黄三七散可明显改善ICH大鼠神经功能缺失症状,显著降低脑系数并减少脑含水量,其对ICH大鼠血清内的NSE、GFAP及Notch1蛋白表达量的升高均有不同程度的抑制作用。ICH大鼠脑血清中NSE的表达与Notch1在mRNA及蛋白水平均呈极显著正相关,而与GFAP的表达在mRNA及蛋白水平均无相关性。而GFAP表达与Notch1正相关。NSE、GFAP及Notch1蛋白的关联性表达研究,表明三者不仅可作为判断急性脑出血损伤发展程度的重要生物标记,还可作为联合选择拮抗蛋白表达的共同药物作用靶点,对急性脑出血的防治具有重要意义。
To study the expression and significance of neuron specific enolase (NSE), glial fibrillary acidic protein (GFAP) and Notch1 receptor protein in neurological deficits of rats with acute cerebral hemorrhage (ICH) induced by Rhubarb Sanqi powder. The rat model of intracerebral hemorrhage was established by heparin collagenase Ⅳ method. The neurological function score, brain coefficient and brain water content, NSE, GFAP and Notch1 mRNA and protein levels in ICH rats before and after administration were investigated. The expression of NSE in ICH rat brain serum had a significant positive correlation with Notch1 mRNA and protein level, but no correlation with GFAP expression at mRNA and protein level. GFAP expression was positively correlated with Notch1.
