目的:探讨抑制转录共激活因子TAZ和M2型丙酮酸激酶(PKM2)在头颈鳞癌(SCCHN)组织的表达及临床意义。方法:收集2014年3月至2020年5月吉林省肿瘤医院手术治疗92例SCCHN患者组织标本,包括25例口腔癌、33例喉癌、l4例下咽癌、6例面颊癌、7例颌面癌、5例舌癌、2例唇癌,另选择25例口腔和咽喉良性病变患者手术时留取的正常口腔、咽喉黏膜组织作为对照组;采用免疫组织化学法检测各组织中TAZ和PKM2的表达水平,并结合临床资料进行n χ2检验。n 结果:TAZ在SCCHN组织中的阳性表达率[83.75%(67/92)],明显高于正常对照组织阳性表达率[54.55%(12/25)],差异有统计学意义(n χ2=10.495,n P<0.01),TAZ表达水平与SCCHN组织学分化程度、美国癌症联合委员会分期(AJCC分期)、淋巴结转移明显相关(n χ2=6.525、5.872、6.107,n P<0.05)。PKM2在SCCHN组织中的阳性表达率[68.48%(63/92)],明显高于正常对照组织阳性表达率[16.00%(4/25)],差异有统计学意义(n χ2=22.122,n P<0.01),PKM2表达水平与SCCHN的AJCC分期、淋巴结转移明显相关(n χ2=6.736、5.312,n P<0.05)。n 结论:TAZ和PKM2在SCCHN组织中的阳性表达率高于对照组织,TAZ和PKM2在SCCHN组织中高表达与肿瘤恶性进展明显相关。“,”Objective:To investigate the expression of transcriptional coactivator with PDZ-binding motif (TAZ) and M2-pyruvate kinase (PKM2) in squamous cell carcinoma of head and neck (SCCHN), and the clinical significance.Methods:The tissue samples of 92 patients with thyroid tumor who underwent surgical resection from March 2014 to May 2020 in Tumor Hospital of Jilin province were collected, including 25 cases of oral carcinoma, 33 cases of laryngeal carcinoma, 14 cases of hypopharyngeal carcinoma, 6 cases of cheek carcinoma, 7 cases of maxillofacial carcinoma, 5 cases of tongue carcinoma and 2 cases of lip carcinoma tissues, and 25 cases of normal oral and pharyngeal mucosal tissues taken during surgery in patients with benign oral and pharyngeal lesions normal tissues served as the control group. Immunohistochemical staining for the paraffin sections (SP method) was used to assess the expression level of TAZ and PKM2 in SCCHN tissues, and the statistical analysis of Chi-square test was carried out in combination with clinical data.Results:The positive rate of TAZ expression was 83.75% (67/92) in SCCHN tissues, higher than 54.55% (12/25) in normal tissues with significant difference (n χ2=10.495, n P<0.01). The expression levels of TAZ were related SCCHN to differentiation degree, American Joint Committee on Cancer clinical stage (AJCC clinical stage) and lymph node metastasis (respectively,n χ2=6.525, 5.872, 6.107, n P<0.05). The positive rate of PKM2 expression was 68.48% (63/92) in SCCHN tissues, higher than 16.00% (4/25) in normal tissues with significant difference (n χ2=22.122, n P<0.01). The expressions level of PKM2 was related SCCHN to AJCC clinical stage and lymph node metastasis (respectively,n χ2=6.736, 5.312, n P<0.05).n Conclusion:The positive rate of TAZ and PKM2 was significantly up-regulated in SCCHN tissues. High expression of TAZ and PKM2 in SCCHN was closely related with malignant progression.
