采用体外培养技术制作人类骨髓基质细胞系,在8cm2平皿中制备原代和传代细胞标本,用ABC法在皿中直接行Vimentin、Desmin、αSmoothMuscleActin单克隆抗体和VonWilebrandFactor抗体染色。结果显示:平皿中细胞分布均匀,无污染、破损和脱片,方法简易,优于盖玻片法,可用于贴壁粘附细胞的免疫组化研究。
Human bone marrow stromal cell lines were made using in vitro culture techniques. Primary and subcultured cell samples were prepared in 8 cm 2 plates and stained with Vimentin, Desmin, α-Smooth Muscle Actin Antibody and Von Wilebrand Factor Antibody using an ABC method in a dish. The results showed that: the cells in the plate were evenly distributed, without pollution, damage and delamination. The method was simple and better than the cover glass method, which could be used to study the immunohistochemistry of adherent cells.
