目的:建立同时测定新疆药桑叶中芦丁、异槲皮苷、槲皮素与绿原酸含量的方法。方法:采用高效毛细管电泳法。以50mmol.L-1硼砂溶液(pH9.6)作为缓冲液,在温度25℃、电压20kV下进行电泳,于254nm波长处进行检测。结果:芦丁、异槲皮苷、绿原酸与槲皮素的检测浓度分别在0.18~105、0.15~100、2.65~169、0.11~79μg.mL-1(r分别为0.9992、0.9989、0.9992、0.9997)范围内与各自峰面积积分值呈良好的线性关系;平均回收率分别为98.4%、97.6%、97.3%、100.3%,RSD分别为1.1%、3.6%、2.1%、2.6%(n=9)。结论:本方法简便、快速、重复性好,结果准确、可靠,可用于桑叶中4种主要黄酮类成分的含量测定和质量检测。
Objective: To establish a method for simultaneous determination of rutin, isoquercitrin, quercetin and chlorogenic acid in medicinal mulberry leaves in Xinjiang. Methods: High performance capillary electrophoresis. A 50 mmol. L-1 borax solution (pH 9.6) was used as a buffer solution and subjected to electrophoresis at a voltage of 20 kV at a temperature of 25 ° C. The detection was performed at a wavelength of 254 nm. Results: The detection concentrations of rutin, isoquercitrin, chlorogenic acid and quercetin were respectively 0.18 ~ 105,0.15 ~ 100,2.65 ~ 169,0.11 ~ 79μg.mL-1 (r = 0.9992,0.9989,0.9992 , 0.9997), respectively. The average recoveries were 98.4%, 97.6%, 97.3% and 100.3%, respectively, and the RSDs were 1.1%, 3.6%, 2.1% and 2.6% = 9). Conclusion: The method is simple, rapid, reproducible, accurate and reliable. It can be used to determine the content and quality of four flavonoids in mulberry leaves.
