【目的】观察益智宁神口服液对抽动—秽语综合征(TS)模型大鼠神经生物学和行为学的影响。【方法】选用SPF级SD大鼠60只,随机分为空白组、模型组、中药组(益智宁神口服液,剂量为2 mL/d)、西药组(氟哌啶醇,剂量为0.15 mg/d)。除空白组外,其他组均采用亚氨基二丙腈(剂量为150 mg.kg-1.d-1)腹腔注射复制TS大鼠模型,造模后第2天开始按设计剂量给药,观察各组大鼠运动行为及刻板运动评分,采用高效液相色谱法检测治疗14 d后各组大鼠脑组织多巴胺(DA)和去甲肾上腺素(NE)的水平。【结果】模型组运动行为、刻板运动积分以及脑组织NE水平均显著升高(P<0.01),DA水平显著降低(P<0.01);中、西药组治疗14 d后均可降低大鼠运动行为、刻板运动积分及脑组织NE水平,升高脑组织DA水平,与模型组比较差异均有显著性意义(P<0.05或P<0.01),提示中药组作用与西药组相仿。【结论】益智宁神口服液治疗TS的作用与其能调整中枢神经递质的水平有关。
【Objective】 To observe the effects of Yizhiningning Oral Liquid on the neurobiology and behavior of Tourette’s syndrome (TS) model rats. 【Methods】 Sixty Sprague-Dawley (SD) SD rats were randomly divided into blank group, model group, Chinese medicine group (Yizhiningsan oral solution, 2 mL / d), western medicine group (haloperidol, 0.15 mg / d). In addition to the blank group, other groups were treated with iminodiacetic acid (dose of 150 mg.kg-1.d-1) intraperitoneal injection of TS rat model, the second day after the model began to be designed dose administration, observed The rats were sacrificed and the motor activity and the score of stereotypia were observed. The levels of dopamine (DA) and norepinephrine (NE) in brain of each group were measured by HPLC after 14 days of treatment. 【Results】 The results showed that motor activity, motor score and brain NE level in model group were significantly increased (P <0.01), while DA level was significantly decreased in model group (P <0.01) (P <0.05 or P <0.01), which suggested that the effect of traditional Chinese medicine group was similar to that of western medicine group. The difference was significant (P <0.05 or P <0.01). 【Conclusion】 The effect of Yizhiningshen oral solution in treating TS is related to its ability to regulate the level of CNS neurotransmitter.
