[目的]建立三倍体毛白杨叶芽再生体系与探索抗生素对叶片再生不定芽的影响。[方法]以叶片为外植体,探索适合三倍体毛白杨遗传转化的再生条件和抗生素对叶片再生不定芽的影响。[结果]结果表明,三倍体毛白杨最适合的不定芽分化培养基配方为MS+6-BA 1.00 mg/L+NAA 0.50 mg/L;在基因工程中,40 mg/L的卡那霉素为选择性抗生素使用浓度,300 mg/L的头孢霉素为抑菌性抗生素使用浓度;再生的不定芽在MS+IBA 0.10 mg/L固体培养基中生根率可达82%。[结论]该研究为毛白杨遗传转化中适宜的培养基成分和抗生素浓度的选择提供科学依据。
[Objective] The research aimed to establish a triploid Populus tomentosa leaf bud regeneration system and explore the effect of antibiotics on adventitious bud regeneration. [Method] With leaves as explants, the regeneration conditions suitable for genetic transformation of Triploid populus tomentosa and the effects of antibiotics on the adventitious bud regeneration of leaves were explored. [Result] The results showed that the most suitable adventitious bud differentiation medium for triploid Populus tomentosa was MS + 6-BA 1.00 mg / L + NAA 0.50 mg / L. In genetic engineering, 40 mg / L kanamycin For the concentration of selective antibiotics, 300 mg / L of cefotaxime was the concentration of antibacterial antibiotics. The regenerated adventitious buds could reach 82% in MS + IBA 0.10 mg / L solid medium. [Conclusion] This study provided a scientific basis for the selection of appropriate medium components and antibiotic concentration in Populus tomentosa for genetic transformation.
