Voltage-gated sodium channels are the basis of neuronal excitation. The voltage-gated sodium channel type 1.8 (Nav 1.8) is a TTX-insensitive sodium channel. In peripheral receptors, Nav1.8 is mainly expressed in nociceptors and a small proportion in non-nociceptors. The study found that patients with congenital fibril neuropathy carry a gain-of-function Nav1.8 mutant. This Nav1.8 mutant is associated with mechanical allodynia in patients with neuropathy. Recently, gain-of-function Nav1.8 mutant mice have been established successfully, laying the foundation for scientists to further study their mechanisms in neuropathic pain.
