用MTT法检测苷甲对K562细胞生长的抑制作用,分别采用形态学方法(光学显微镜、倒置显微镜、荧光显微镜)检查在苷甲作用下K562细胞形态的变化以及用流式细胞术观察在苷甲作用下K562细胞凋亡及周期变化的情况.结果显示:苷甲抑制K562细胞的生长,其24、48、72h的IC50值分别为19.7、18.5、15.9μmol/L.苷甲诱导K562细胞出现典型凋亡细胞的形态学特征.20μmol/L苷甲作用K562细胞6 h,流式细胞仪即检出亚倍体峰,G2/M期细胞增多.随着作用时间延长,细胞被阻滞于G2/M期更加明显,细胞的凋亡率也增加.该实验证明:苷甲能抑制K562细胞的生长,把细胞阻滞在G2/M期并诱导细胞发生凋亡.
The inhibitory effect of glycosides on the growth of K562 cells was detected by MTT assay. The morphological changes of K562 cells were observed by morphological methods (light microscope, inverted microscope and fluorescence microscope), and the morphological changes of K562 cells were observed by flow cytometry. The results showed that glycoside inhibited the growth of K562 cells at 24,48,72h and IC50 values were 19.7,18.5 and 15.9μmol / Morphological characteristics of apoptotic cells. K562 cells treated with 20 μmol / L of glycoside for 6 h, the cells were detected by flow cytometry and the number of cells was increased at G2 / M phase. The cells were arrested at G2 / M phase more obvious, the cell apoptosis rate also increased.This experiment shows that: Glycoside can inhibit the growth of K562 cells, the cells arrested in the G2 / M phase and induce cell apoptosis.
