目的 :探讨α 突触核蛋白的非淀粉样成分 (NAC)对多巴胺能细胞的毒性作用。方法 :PC12细胞经不同浓度的NAC处理 48h后 ,观察细胞形态改变 ,MTT法测定细胞活力 ;TUNEL和Annexin V法流式细胞仪检测调亡 ;硫磺素S染色研究蛋白聚集。结果 :NAC处理 48h后 ,PC12细胞突起样结构消失 ;与对照组比较 ,NAC >2 5 μmol·L使细胞活力明显下降 (P <0 0 5 ) ,并诱导出现凋亡及蛋白聚集。结论 :ANC对多巴胺能细胞有毒性作用 ,并引起蛋白聚集。
Objective: To investigate the toxic effects of α-synuclein non-amyloid (NAC) on dopaminergic cells. Methods: PC12 cells were treated with different concentrations of NAC for 48h, the morphological changes of cells were observed, and cell viability was measured by MTT assay. TUNEL and Annexin V were detected by flow cytometry. Thioflavin S staining was used to study the protein aggregation. RESULTS: After 48 h of NAC treatment, the neurite outgrowth in PC12 cells was observed. Compared with the control group, NAC> 25 μmol·L significantly decreased cell viability (P <0.05) and induced apoptosis and protein accumulation. Conclusion: ANC has a toxic effect on dopaminergic cells and causes protein aggregation.
