目的研究地黄饮子孵育骨髓间充质干细胞(bone marrow stromal stem cells,BMSCs)移植对脑梗死大鼠脑组织中基质金属蛋白酶-9(MMP-9)和金属蛋白酶组织抑制剂-1(TIMP-1)的影响,探讨其对大脑的保护作用。方法将健康SPF级Wistar大鼠随机分为假手术组、模型组、BMSCs组、维甲酸组和地黄饮子组,用线栓法制成Wistar大鼠大脑中动脉缺血模型(MCAO),术后24 h假手术组、模型组大鼠经尾静脉注射生理盐水,其余各组大鼠经尾静脉输入相应药物孵育的BMSCs。移植后7 d,14 d断头取脑,用免疫组化法检测脑组织中MMP-9和TIMP-1的表达情况。结果与假手术组比较,模型组MMP-9表达明显增加(P<0.05);地黄饮子组、维甲酸组、BMSCs组与模型组比较,MMP-9的表达均降低(P<0.05),但仍高于假手术组。模型组TIMP-1的表达明显降低(P<0.05),各治疗组均增高(P<0.05)。结论地黄饮子孵育BMSCs可通过调节MMP-9与TIMP-1的表达,进而发挥脑保护作用。
Objective To study the effects of Rehmannia glutinosa submerged in bone marrow stromal stem cells (BMSCs) transplantation on the expression of matrix metalloproteinase-9 (MMP-9) and tissue inhibitor of metalloproteinase-1 (TIMP- 1), to explore its protective effect on the brain. Methods Wistar rats were randomly divided into sham operation group, model group, BMSCs group, retinoic acid group and Rehmanniae Decoction group. MCAO was induced by Wistar rats by thread method. After operation After 24 h sham operation, rats in the model group were injected with saline through the caudal vein, and rats in other groups were injected with BMSCs via corresponding tail vein. At 7 and 14 days after transplantation, the brain was decapitated and the expression of MMP-9 and TIMP-1 in the brain tissue was detected by immunohistochemistry. Results Compared with the sham operation group, the expression of MMP-9 in the model group was significantly increased (P <0.05). Compared with the model group, the expression of MMP-9 in the Decoction group, the retinoic acid group and the BMSCs group were significantly decreased (P <0.05) But still higher than sham group. The expression of TIMP-1 in the model group was significantly lower (P <0.05), and all the treatment groups were higher (P <0.05). Conclusion Rehmanniae tuber incubation of BMSCs can exert cerebral protective effect by regulating the expression of MMP-9 and TIMP-1.
