目的研究人脑星形细胞肿瘤中FHIT、PCNA蛋白的差异表达,探讨它们与病理分级之间的关系。方法应用免疫组化SP法检测了50例星形细胞肿瘤中不同级别的FHIT、PCNA蛋白的表达水平,以10例非肿瘤脑组织作对照。结果非肿瘤脑组织FHIT、PCNA蛋白阳性表达率分别为100%,0%,星形细胞肿瘤中FHIT、PCNA蛋白阳性表达率分别为40%,86%;尽管在星形细胞肿瘤中FHIT、PCNA蛋白表达总体差异有统计学意义(P<0.05),但Ⅲ级与Ⅳ级比较差异无统计学意义(P>0.05);经统计学分析FHIT、PCNA蛋白呈显著负相关(P<0.01)。结论FHIT、PCNA蛋白的表达可能与星形细胞肿瘤的恶性程序有关,星形细胞肿瘤中FHIT、PCNA蛋白表达之间的显著负相关,提示FHIT蛋白可能对细胞增殖具有负性调控作用。
Objective To study the differential expression of FHIT and PCNA proteins in human brain astrocytic tumors and to explore their relationship with pathological grade. Methods Immunohistochemical SP method was used to detect the expression levels of FHIT and PCNA protein in 50 cases of astrocytic tumors with 10 cases of non-tumor brain tissue as control. Results The positive expression rates of FHIT and PCNA protein in non-tumor brain tissue were 100% and 0%, respectively. The positive rates of FHIT and PCNA protein in astrocytic tumors were 40% and 86%, respectively. Although FHIT and PCNA (P <0.05), but there was no significant difference between grade Ⅲ and grade Ⅳ (P> 0.05). There was a significant negative correlation between FHIT and PCNA <0.01). Conclusions The expression of FHIT and PCNA may be related to the malignant process of astrocytoma. The negative correlation between the expression of FHIT and PCNA in astrocytic tumors suggests that FHIT may negatively regulate cell proliferation.
