目的:探讨缺血后处理对大鼠缺血再灌注损伤后CD105及endostatin表达的影响。方法:将雄性Wistar大鼠随机分为三组,分别为假手术组(Sham组)、缺血再灌注损伤组(IRI组)及缺血后处理组(IPO组)。采用全自动生化分析仪检测血清尿素氮(BUN)及肌酐(Cr)含量;用HE染色观察病理组织学变化;用免疫组织化学法和蛋白质印迹法(Western blot)检测肾组织中CD105及endostatin的表达。结果:IRI组血清BUN及Cr含量较Sham组明显升高,IPO组血清BUN及Cr含量介于Sham组和IRI组之间;HE染色检测发现IRI组肾小管上皮细胞坏死明显;肾小管管腔扩张,IPO组较IRI组肾小管上皮坏死明显减轻;免疫组织化学以及Western blot检测均发现CD105在Sham组表达最强,IRI组表达明显减弱,IPO组表达则介于前二者之间;endostatin在Sham组表达最弱,在IRI组最强,IPO组则稍强于Sham组而明显弱于IRI组。结论:缺血后处理能减弱缺血再灌注损伤过程中endostatin的表达量,同时增加CD105的表达,因而有助于减轻肾脏的缺血再灌注损伤。
Objective: To investigate the effect of ischemic postconditioning on the expression of CD105 and endostatin after ischemia-reperfusion injury in rats. Methods: Male Wistar rats were randomly divided into three groups: sham group, IRI group and IPO group. Serum urea nitrogen (BUN) and creatinine (Cr) were detected by automatic biochemical analyzer. The histopathological changes were observed by HE staining. The expressions of CD105 and endostatin in renal tissues were detected by immunohistochemistry and Western blotting expression. Results: Serum levels of BUN and Cr in IRI group were significantly higher than those in Sham group. Serum levels of BUN and Cr in IPO group were between Sham group and IRI group; necrosis of renal tubular epithelial cells in IRI group was found by HE staining; The expression of CD105 in Sham group and IRI group were significantly lower than those in IRI group. Immunohistochemistry and Western blot showed that the expression of CD105 was the highest in Sham group and significantly decreased in IRI group. The expression of endostatin The expression was the weakest in Sham group, strongest in IRI group, and slightly weaker than that in IRI group in IPO group. Conclusion: Ischemic postconditioning can reduce the expression of endostatin during ischemia-reperfusion injury and increase the expression of CD105, which may help to reduce renal ischemia-reperfusion injury.
