目的:研究p53蛋白,PCNA,DNA含量在宫颈癌及癌前病变组织中的表达及其意义.方法:用免疫组化ABC法和图像分析法,检测p53蛋白、PCNA和DNA含量,对56例正常宫颈粘膜、宫颈上皮内瘤变、宫颈浸润性鳞癌进行研究.结果:p53表达阳性率、PCNA细胞增殖指数和DNA含量随癌组织分级依次增高,p53蛋白在正常粘膜不表达,在宫颈上皮瘤变和原位癌少量表达,在宫颈浸润癌表达明显增高.结论:p53含量和细胞活性增高与宫颈粘膜癌变发生及恶性程度有关.PCNA增殖指数、DNA含量和多倍体细胞的出现,对宫颈癌的早期诊断有一定意义.p53蛋白的阳性表达在宫颈癌发生中不是一个晚期事件.
Objective: To study the expression and significance of p53, PCNA and DNA in cervical cancer and precancerous lesions. Methods: Immunohistochemical ABC method and image analysis were used to detect the expression of p53, PCNA and DNA in 56 cases of normal cervical mucosa, cervical intraepithelial neoplasia and cervical squamous cell carcinoma. Results: The positive rate of p53, the proliferation index of PCNA cells and DNA content increased with the order of the cancerous tissues. The p53 protein was not expressed in normal mucosa, but slightly expressed in cervical epithelial neoplasia and carcinoma in situ, and significantly increased in invasive cervical carcinoma. Conclusion: The increase of p53 and cell activity is related to the carcinogenesis and malignancy of cervical mucosa. Proliferation index of PCNA, DNA content and the appearance of polyploid cells have certain significance for the early diagnosis of cervical cancer. The positive expression of p53 protein is not a late event in the development of cervical cancer.
