目的从分子水平研究了诺氟沙星金属配合物与DNA的相互作用,为进一步开发其药用价值及其金属配合物的抗肿瘤活性提供一定的理论基础。方法合成新的诺氟沙星-饵金属配合物,应用熔点测定,摩尔电导率测定,IR谱,H1NMR谱,元素分析和质谱分析系等手段对所合成的配合物进行了表征。并应用荧光、黏度和热变性等方法,对此配合物与小牛胸腺DNA作用方式进行了研究。结论配合物与DNA作用时,荧光发生淬灭,黏度增大,热变性温度升高;Scatchard图表明配合物对溴化乙锭(EB)与DNA的结合为竞争性抑制。实验结果一致表明,配合物与DNA作用方式为嵌插结合。
Objective To study the interaction between Norfloxacin metal complexes and DNA at molecular level and to provide some theoretical basis for further development of its medicinal value and anti-tumor activity of its metal complexes. Methods Novel norfloxacin - bait metal complexes were synthesized and their structures were characterized by means of melting point, molar conductivity, IR, HNMR, elemental analysis and mass spectrometry. Fluorescence, viscosity and thermal denaturation methods were used to study the interaction between the complex and calf thymus DNA. Conclusions When the complex interacts with DNA, the fluorescence quenches, the viscosity increases, and the thermal denaturation temperature increases. The Scatchard plot shows that the complex inhibits the binding of ethidium bromide (EB) to DNA. The experimental results show that the interaction between the complex and DNA is intercalated.
