目的观察骨髓间充质干细胞(BMSC)对肺气肿模型大鼠炎症因子和肺组织病理学变化的影响,探讨肺气肿的发病机制。方法采集3只SD幼鼠骨髓细胞,培养传代;将40只雄性SD大鼠随机分成对照、肺气肿模型组、高、中、低浓度BMSC组,建立肺气肿模型,大鼠尾静脉注射不同浓度的BMSC治疗。观察肺组织病理学变化及肺泡灌洗液细胞计数,用ELISA检测各组大鼠BALF中IL-17和IL-6水平。结果肺气肿模型组和3个BMSC组BALF中白细胞总数、巨噬细胞数及中性粒细胞百分比均高于对照组(P<0.05)。与正常组比较,3个BMSC组、肺气肿模型组BALF和血清中IL-17、IL-6水平明显升高(P<0.05)。BMSC组、肺气肿模型组与正常组比较,MLI增加,MAN减少(P<0.05)。结论BMSC能改善肺气肿大鼠模型的病理改变,且浓度越高疗效越好。
Objective To observe the effects of bone marrow mesenchymal stem cells (BMSCs) on the inflammatory cytokines and pathological changes in rats with emphysema and to explore the pathogenesis of emphysema. Methods Bone marrow cells from 3 SD rats were collected and passaged. 40 male Sprague-Dawley rats were randomly divided into control group, emphysema model group, high, medium and low concentration BMSC group. The model of emphysema was established. The tail vein Different concentrations of BMSC treatment. The histopathological changes of lung tissue and the cell counts of BALF were observed. The levels of IL-17 and IL-6 in BALF of rats in each group were detected by ELISA. Results The total number of leucocytes, the number of macrophages and the percentage of neutrophils in BALF of emphysema model group and 3 BMSC group were higher than those of control group (P <0.05). Compared with the normal group, the levels of IL-17 and IL-6 in the BALF and serum of the three BMSC and emphysema models were significantly increased (P <0.05). In BMSC group and emphysema model group, MLI increased and MAN decreased (P <0.05) compared with normal group. Conclusion BMSC can improve the pathological changes of rat model of emphysema, and the higher the concentration, the better the curative effect.
