目的探讨氯化钴(CoCl2)诱发缺氧对ECV-304内皮细胞低氧诱导因子HIF-1α表达及细胞定位的影响。方法采用western blotting方法检测CoCl2对HIF-1α表达的影响,并通过免疫荧光方法检测HIF-1α在ECV-304细胞系中的表达,并观察CoCl2对HIF-1α细胞定位的影响。结果 ECV-304内皮细胞中低氧诱导因子HIF-1α在细胞质表达,CoCl2诱发缺氧可上调细胞中HIF-1α的表达,并且促进HIF-1α入核。结论 CoCl2诱发缺氧可上调具有内皮细胞特性的ECV-304细胞中HIF-1α的表达及入核。
Objective To investigate the effects of cobalt chloride (CoCl 2) -induced hypoxia on hypoxia-inducible factor 1 (HIF-1α) expression and cell location in ECV-304 endothelial cells. Methods The effect of CoCl2 on the expression of HIF-1α was detected by western blotting. The expression of HIF-1α in ECV-304 cell line was detected by immunofluorescence staining and the effect of CoCl2 on the localization of HIF-1α cells. Results The hypoxia-inducible factor HIF-1α was expressed in the cytoplasm of ECV-304 endothelial cells. CoCl2-induced hypoxia increased the expression of HIF-1α and promoted the expression of HIF-1α. Conclusion CoCl2-induced hypoxia can up-regulate the expression of HIF-1α and enter into the nucleus in ECV-304 cells with endothelial cell characteristics.
