目的观察红景天注射液对心肺复苏后大鼠心脏的保护作用。方法 56只SD成年健康大鼠随机分为7组,每组8只。窒息法建立大鼠心肺复苏模型,在不同时间点给予红景天注射液(复苏前,复苏同时,复苏开始5min、30min、1h),并设立假手术组和不给药组作为对照。自主循环恢复维持2h后检测心肌组织白细胞介素6(interleukin 6,IL-6)、肿瘤坏死因子α(tumor necrosis factor alpha,TNF-α)以及血清肌钙蛋白T(cardiac troponin T,cTnT)水平,常规苏木精-伊红(hematoxylin eosin,HE)染色病理观察。结果红景天呈时间依赖性降低心肺复苏后心肌组织IL-6、TNF-α以及血清cTnT水平(P<0.05),病理观察显示复苏前给药组心肌损伤最轻(P<0.05),复苏1h给药与不给药相比无明显差异。结论红景天可有效保护心肺复苏后大鼠心肌,作用呈时间依赖性,提前给药作用最明显,复苏1h给药基本无效。
Objective To observe the protective effect of Rhodiola on the rat heart after cardiopulmonary resuscitation. Methods 56 SD adult healthy rats were randomly divided into 7 groups with 8 rats in each group. Asphyxia method was used to establish rat model of cardiopulmonary resuscitation (CPR). Rhodiola was injected at different time points (before resuscitation, resuscitation, resuscitation 5min, 30min, 1h) and sham-operation group and non-administration group as control. The levels of interleukin 6 (IL-6), tumor necrosis factor alpha (TNF-α) and cardiac troponin T (cTnT) , Routine hematoxylin - eosin (HE) staining pathological observation. Results Rhodiola rose in a time-dependent manner to reduce myocardial IL-6, TNF-α and serum cTnT levels after cardiopulmonary resuscitation (P <0.05). Pathological examination showed that myocardial damage was the lightest before resuscitation (P <0.05) 1h administration and no administration compared to no significant difference. Conclusion Rhodiola can effectively protect myocardium after cardiopulmonary resuscitation in a time-dependent manner, with the most obvious effect being given in advance. The administration of rhodiola at 1 hour after resuscitation is basically ineffective.
