目的探讨EZH2在肝细胞癌(肝癌)中的表达及其临床意义。方法采用RT-PCR、免疫组织化学和Western blotting方法分析肝癌及癌旁组织EZH2mRNA转录和蛋白表达水平及其与临床病理因素的关系。结果在42例肝癌组织中检出26例EZH2mRNA,而在相应的42例癌旁组织中只检出12例,癌组织EZH2mRNA阳性率高于癌旁组织,差异有显著性(P<0.01)。免疫组织化学结果显示,42例肝癌组织中有25例呈EZH2蛋白表达阳性,相应的42例癌旁组织中只有9例呈阳性,癌组织和癌旁组织蛋白表达率有显著性差异(P<0.01)。取上述42例中的18例肝癌及癌旁组织做Western blotting,结果显示,18例肝癌及癌旁组织均有EZH2蛋白表达,癌组织EZH2表达量明显高于癌旁组织(0.90±0.30和0.36±0.21,P<0.01)。EZH2与临床病理特征分析中,EZH2mRNA在低分化组的表达明显高于高-中分化组,而与肿瘤直径、肝内外转移无相关性。但EZH2蛋白表达与肿瘤直径、分化程度、肝内外转移之间均无相关性。结论 EZH2在肝癌组织中表达上调,提示EZH2在肝癌的发生发展中起重要作用。
Objective To investigate the expression of EZH2 in hepatocellular carcinoma (HCC) and its clinical significance. Methods RT-PCR, immunohistochemistry and Western blotting were used to analyze the mRNA and protein expression level of EZH2 in hepatocellular carcinoma (HCC) and adjacent non-cancerous tissues and their relationship with clinicopathological parameters. Results Twenty-six EZH2 mRNAs were detected in 42 HCC tissues, while only 12 were detected in the corresponding 42 paracancerous tissues. The positive rate of EZH2 mRNA in cancer tissues was significantly higher than that in paracancerous tissues (P <0.01). The results of immunohistochemistry showed that EZH2 protein was positive in 25 of 42 HCC tissues, and only 9 of 42 paracancer tissues were positive. There was significant difference in the expression of protein between cancer tissues and paracancerous tissues (P < 0.01). Western blotting was performed on 18 cases of HCC and paracancerous tissue from the above 42 cases. The results showed that EZH2 protein was expressed in 18 cases of HCC and paracancerous tissues, and the expression of EZH2 in cancer tissues was significantly higher than that in paracancerous tissues (0.90 ± 0.30 and 0.36 ± 0.21, P <0.01). In EZH2 and clinicopathological analysis, the expression of EZH2mRNA in poorly differentiated group was significantly higher than that in high-moderately differentiated group, but not correlated with tumor diameter and intrahepatic metastasis. However, there was no correlation between EZH2 protein expression and tumor diameter, differentiation degree and intrahepatic metastasis. Conclusion EZH2 is up-regulated in HCC tissues, suggesting that EZH2 plays an important role in the development of HCC.
