目的:建立HPLC-PDA同时测定白术中白术内酯Ⅰ和白术内酯Ⅲ含量的方法,比较不同温度鼓风烘制与硫熏白术中这2种成分的含量,探究代替硫磺熏蒸的最佳温度鼓风烘制保存的加工方法。方法:不同温度鼓风烘制和硫磺熏蒸的白术提取物的分析采用InertsilODS-SP色谱柱,流速1.0 m L·min~(-1),流动相乙腈-水(52∶48),检测波长220 nm。结果:白术内酯Ⅰ和白术内酯Ⅲ的线性范围分别为20.4~326.4,20.9~334.4 ng(r>0.999 5),加样回收率分别为99.43%和99.20%,RSD分别为0.2%和0.3%。70℃烘制的白术中白术内酯Ⅰ和白术内酯Ⅲ的含量最高。结论:高温鼓风烘制及硫磺熏蒸均会减少白术内酯Ⅰ和白术内酯Ⅲ的含量,建议采用70℃鼓风烘制法替代硫磺熏蒸作为白术的加工方法,该方法便捷、可靠、安全,为白术的质量控制提供参考。
OBJECTIVE: To establish a HPLC-PDA method for simultaneous determination of atractyloside I and atractylenolide III in Atractylodes macrocephalae and to compare the contents of these two components in blast furnace and sulfur smoked aphrodisiac at different temperatures and explore the optimal temperature to replace sulfur fumigation Blow drying preservation of processing methods. Methods: The Anemiasil ODS column was used to analyze the extracts of Atractylodes macrocephala at different temperatures by air-blowing and sulfur fumigation. The flow rate was 1.0 m L · min -1 and the mobile phase was acetonitrile-water (52:48) Wavelength 220 nm. Results: The linear ranges of atractyloside Ⅰ and atractyloside Ⅲ were 20.4 ~ 326.4 and 20.9 ~ 334.4 ng respectively (r> 0.999 5), the recoveries were 99.43% and 99.20% respectively, and the RSDs were 0.2% and 0.3% %. The content of atractyloside Ⅰ and atractyloside Ⅲ in Atractylodes macrocephala which was baked at 70 ℃ was the highest. CONCLUSION: High-temperature blast drying and sulfur fumigation both reduce the content of atractylenolide I and atractylenolide III. It is suggested that the method of blowing fumigation at 70 ℃ instead of sulfur fumigation is the processing method of Atractylodes macrocephala which is convenient, reliable and safe. , Provide a reference for the quality control of Atractylodes.
