目的:研究生存素(survivin)反义寡核苷酸(antisense oligodeoxynucleotide,ASODN)对人乳腺癌MCF-7细胞株基质金属蛋白酶(matrix metalloproteinase,MMP)-2及MMP-9表达的影响,以进一步阐明生存素介导乳腺癌浸润及转移的机制。方法:经脂质体介导将生存素ASODN转染人乳腺癌细胞株MCF-7,用荧光显微镜检测转染效率,用逆转录聚合酶链反应(RT-PCR)、蛋白质印迹法检测转染细胞生存素、MMP-2及MMP-9表达改变。结果:荧光显微镜检测结果显示生存素ASODN可成功转染MCF-7细胞。与空脂质体组、空ASODN组相比,转染组细胞生存素、MMP-2、MMP-9 mRNA和蛋白的表达均显著降低。结论:生存素ASODN转染可有效抑制MCF-7细胞生存素基因的表达,并降低其MMP-2、MMP-9表达,可能对乳腺癌具有基因治疗的效果。
Objective: To investigate the effect of survivin antisense oligodeoxynucleotide (ASODN) on the expression of matrix metalloproteinase (MMP) -2 and MMP-9 in human breast cancer cell line MCF-7, To elucidate the mechanism of survivin-mediated breast cancer invasion and metastasis. METHODS: Survivin ASODN was transfected into human breast cancer cell line MCF-7 by liposome-mediated transfection. The transfection efficiency was detected by fluorescence microscopy. The transfection efficiency was detected by reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blotting Cell Survivin, MMP-2 and MMP-9 expression changes. Results: Fluorescence microscopy showed that survivin ASODN could be successfully transfected into MCF-7 cells. Compared with empty liposome group and empty ASODN group, the expressions of survivin, MMP-2, MMP-9 mRNA and protein in transfected group were significantly decreased. CONCLUSIONS: Survivin ASODN transfection can effectively inhibit the expression of survivin gene and decrease the expression of MMP-2 and MMP-9 in MCF-7 cells, which may have the effect of gene therapy on breast cancer.
