目的:观察金钗石斛菲醌对人卵巢浆液性囊腺癌细胞系HO-8910PM的增生和转移能力的影响,检测其对卵巢癌增生和转移相关基因CASP3,CASP9,CAV-1和SOX2的分子水平表达,从而研究金钗石斛菲醌在细胞和分子水平对卵巢癌的治疗作用以及其相关分子机制。方法:运用噻唑蓝还原法(MMT)方法来检测金钗石斛菲醌对人卵巢癌细胞HO-8910PM的抗增殖作用,而同时运用Transwell法来检测药物对于细胞的转移能力的作用变化。再通过逆转录聚合酶链式反应和蛋白质印迹的实验来检测HO-8910PM细胞中凋亡与转移相关基因的表达变化以及蛋白水平的变化。结果:实验结果证明金钗石斛菲醌对人卵巢癌细胞具有抗侵袭以及转移的治疗作用。MTT检测结果显示金钗石斛菲醌在3μmol/L和10μmol/L对于卵巢癌细胞的抑增殖作用显著,而transwell法的结果也表明了其在3μmol/L和10μmol/L对癌细胞的抗转移能力。其治疗效果与阳性顺铂10μmol/L治疗组在统计学意义上相近。而mRNA,蛋白水平测定,表明金钗石斛菲醌在3μmol/L对人卵巢癌细胞通过上调CASP3,CASP9,CAV1和下调SOX2的表达,从而调节相关信号通路,最终在分子水平证明金钗石斛菲醌对人卵巢癌细胞增殖和转移的抑制作用。结论:金钗石斛菲醌通过上调CASP3,CASP9,CAV1和下调SOX2的表达,进而抑制人卵巢癌细胞的增殖和转移。
OBJECTIVE: To observe the effect of Dendrobium nobile Lindmannin on the proliferation and metastasis of human ovarian serous cystadenocarcinoma cell line HO-8910PM and to detect the expression of CASP3, CASP9, CAV-1 and SOX2 in ovarian cancer So as to study the therapeutic effect of Dendrobium mofetili at the cellular and molecular level on ovarian cancer and its related molecular mechanisms. Methods: The antiproliferative effect of Dendrobium nobile Lindl109 on human ovarian cancer cells HO-8910PM was detected by thiazolyl blue reduction method (MMT). Transwell assay was used to detect the effect of drugs on cell migration. The changes of HO-8910PM cells apoptosis-related genes and protein levels were detected by RT-PCR and Western blotting. Results: The experimental results show that the anti-invasive and metastatic therapeutic effect of Dendrobium nobilein on human ovarian cancer cells. MTT assay showed that 3μmol / L and 10μmol / L of doxorubicin showed significant inhibitory effects on ovarian cancer cells, while the results of transwell assay also showed that the anti-metastatic effect of 3μmol / L and 10μmol / L on cancer cells ability. Its therapeutic effect and positive cisplatin 10μmol / L treatment group was statistically significant. However, the mRNA and protein levels of Dendrobium nobile Lindl had no effect on the expression of CASP3, CASP9, CAV1 and down-regulated the expression of SOX2 in 3μmol / L human ovarian cancer cells, Inhibition of Quinoids on Proliferation and Metastasis of Human Ovarian Cancer Cells. Conclusion: Dendrobium nobilein can inhibit the proliferation and metastasis of human ovarian cancer cells by up-regulating CASP3, CASP9, CAV1 and down-regulating the expression of SOX2.
