目的 用HPLC检测人血清中的特布他林浓度。方法 样品用硅胶萃取小柱纯化浓集后用C8柱 (5 μm ,2 5 0mm× 4.6mm)作分析柱 ,用硫酸沙丁胺醇作内标 ,以pH2 .8,0 .0 3mol·L- 1 NH4H2 PO4缓冲液 CH3OH CH3CN(90∶3∶7)为流动相 ,在波长为2 2 4nm处紫外检测。结果 标准曲线在 0 .5~ 32 μg·L- 1 范围内有良好线性关系 (r =0 .9998) ,最低检测浓度为 0 .2 μg·L- 1 ,平均方法回收率为 (10 0 .7± 3 .1) % ,日内RSD小于 3.7% ,日间RSD小于 7.5 %。结论 方法简便 ,快速准确 ,适宜血清中特布他林的测定
Objective To detect the concentration of terbutaline in human serum by HPLC. Methods The sample was purified by silica gel column and concentrated with C8 column (5 μm, 250 mm × 4.6 mm) for analysis. Salbutamol sulfate was used as internal standard and pH2 .8,0. 3mol·L-1 NH4H2 PO4 buffer CH3OH CH3CN (90: 3: 7) as the mobile phase, at a wavelength of 224nm UV detection. Results The calibration curve showed a good linearity (r = 0.9998) in the range of 0.5 ~ 32 μg · L-1 and the lowest detection limit was 0.2 μg · L-1. The average recovery was 10%. 7 ± 3 .1)%, intraday RSD less than 3.7%, daytime RSD less than 7.5%. Conclusion The method is simple, rapid and accurate, suitable for the determination of terbutaline in serum
