目的:探讨nm23-H1转染对肺癌细胞钙调素和基质金属蛋白酶表达的影响。方法:利用半定量RT-PCR技术检测转染前后E-cadherin、MMP-9基因的mRNA表达;利用流式细胞仪技术检测转染前后Ecadherin、MMP-9基因的蛋白表达。结果:转染后细胞株E-cadherin mRNA与蛋白表达上调,转染后细胞株MMP-9 mRNA与蛋白表达下调。结论:nm23-H1基因对肺癌细胞株L9981 E-cadherin mRNA与蛋白表达具有正调控作用,对MMP-9 mRNA与蛋白表达具有负调控作用。
Objective: To investigate the effect of nm23-H1 transfection on the expression of calmodulin and matrix metalloproteinase in lung cancer cells. Methods: The mRNA expressions of E-cadherin and MMP-9 before and after transfection were detected by semi-quantitative RT-PCR. The protein expressions of Ecadherin and MMP-9 before and after transfection were detected by flow cytometry. Results: After transfection, the expression of E-cadherin mRNA and protein were up-regulated, while the expression of MMP-9 mRNA and protein was down-regulated after transfection. CONCLUSION: The nm23-H1 gene has a positive regulatory effect on the expression of E-cadherin mRNA and protein in lung cancer cell line L9981, and negatively regulates the expression of MMP-9 mRNA and protein.
