目的研究EMP1在喉癌中的作用。方法选取51例喉癌标本及其癌旁组织,构建EMP1过表达载体,转染人喉癌Hep-2细胞系,应用MTT法、克隆形成实验、流式细胞术及Transwell法分别检测细胞的增殖、凋亡及迁移情况,明确EMP1过表达后对喉癌细胞产生的生物学效应。结果 EMP1过表达后,Hep2细胞的增殖能力明显减弱,细胞克隆形成数目平均值明显减少,细胞的凋亡率平均值明显上调,穿过小室滤膜的细胞数平均值明显下调。即喉癌细胞的增殖能力及迁移能力减弱,凋亡增强。结论 EMP1基因在喉癌的发生发展中发挥抑癌基因的作用。
Objective To investigate the role of EMP1 in laryngeal cancer. Methods Totally 51 specimens of laryngeal cancer and their adjacent tissues were selected to construct EMP1 overexpression vector and transfected into human laryngeal carcinoma Hep-2 cell line. MTT assay, clone formation assay, flow cytometry and Transwell assay were used to detect the cell proliferation , Apoptosis and migration, clarify the biological effects of EMP1 overexpression on laryngeal carcinoma cells. Results After EMP1 overexpression, the proliferation of Hep2 cells was significantly weakened, the average number of cell clone formation was significantly decreased, the average apoptotic rate of cells was significantly up-regulated, and the average number of cells passing through the cell membrane filter was significantly down-regulated. Laryngeal cancer cell proliferation and migration ability weakened, apoptosis increased. Conclusion EMP1 gene plays a role of tumor suppressor gene in the development of laryngeal carcinoma.
