目的研究HERC4在人乳腺癌组织中的表达,探讨其与乳腺癌临床特征的相关性。方法 RT-qPCR法检测67例乳腺癌组织及配对癌旁乳腺组织标本中HERC4的mRNA表达水平;Western blot和免疫组化法检测67例乳腺癌组织及配对癌旁乳腺组织标本中HERC4的蛋白表达水平。结果 RT-qPCR结果显示HERC4的mRNA的表达水平在乳腺癌组织中较癌旁乳腺组织高(P<0.05)。Western blot结果显示HERC4蛋白在乳腺癌组织中高表达(P<0.05)。免疫组化结果表明HERC4主要在细胞质中表达。乳腺癌组织中HERC4阳性表达率为61.2%,癌旁乳腺组织阳性表达率为19.4%。HERC4的表达与乳腺癌TNM分期和组织学分级密切相关(P<0.05)。结论 HERC4与乳腺癌的发生和发展存在相关关系,并有可能作为乳腺癌早期诊断标志物和治疗靶点。
Objective To study the expression of HERC4 in human breast cancer and explore its correlation with clinical features of breast cancer. Methods RT-qPCR was used to detect the mRNA expression of HERC4 in 67 cases of breast cancer tissues and paired para-cancerous breast tissues. Western blot and immunohistochemistry were used to detect the protein expression of HERC4 in 67 cases of breast cancer tissues and matched paracancerous breast tissues Level. Results The RT-qPCR results showed that the mRNA expression level of HERC4 was higher in breast cancer tissues than in paracancerous breast tissues (P <0.05). Western blot results showed that HERC4 protein was highly expressed in breast cancer tissues (P <0.05). Immunohistochemistry showed that HERC4 was mainly expressed in the cytoplasm. The positive rate of HERC4 expression in breast cancer tissues was 61.2%, while the positive expression rate of breast cancer tissues in adjacent tissues was 19.4%. The expression of HERC4 was closely related to TNM staging and histological grade of breast cancer (P <0.05). Conclusion HERC4 is correlated with the occurrence and development of breast cancer and may serve as an early diagnosis marker and target of breast cancer.
