目的:建立RP-HPLC测定桂郁金中吉玛酮的含量。方法:采用正交试验优化样品处理的提取条件,采用RP-HPLC检测吉玛酮的含量,Phenomenex C18色谱柱(4.6 mm×250 mm,5μm),流动相76%乙腈,流速1.0 mL.min-1,检测波长为217 nm。结果:吉玛酮在0.056 7~2.324 7μg呈良好的线性关系(r=0.999 99),平均回收率98.05%,RSD 0.69%;容县产桂郁金吉玛酮含量最高,而平南产含量最低;桂郁金块根炮制品及桂郁金茎叶中吉玛酮的含量明显低于自然阴干的桂郁金块根。结论:吉玛酮含量测定方法简单、准确、快速;不同产地、不同部位以及不同炮制品的桂郁金吉玛酮含量相差很大。
Objective: To establish an RP-HPLC method for the determination of jimarmazuril in Guiyujin. Methods: The extraction conditions were optimized by orthogonal test. The content of jimaone was determined by RP-HPLC. Phenomenex C18 column (4.6 mm × 250 mm, 5 μm) with mobile phase of 76% acetonitrile and flow rate of 1.0 mL · min- 1, detection wavelength of 217 nm. Results: The results showed that there was a good linear correlation between jimaprone and 0.056 7 ~ 2.324 7μg (r = 0.999 99), the average recovery was 98.05% and RSD 0.69% ; Guiyu gold root canned products and Guiyu gold leaf jimalei content was significantly lower than the natural shade of Guiyu gold roots. Conclusion: The method of determination of jimaone is simple, accurate and rapid. The content of guanyujinjimane in different areas, different parts and different processed products varies greatly.
