本文报告用骨髓细胞及血细胞短期培养法对6例恶性网状细胞病患者的细胞分裂中期染色体进行观察的结果。细胞培养方法基本上按照项维等报导的短期培养法略加修改。培养基系用含有0.5％水解乳蛋白的Earle液,加入AB型人血清或初生小牛血清及植物血凝集素(PHA)。培养时间72小时,培养终止前4小时,加入秋水仙素,使每毫升培养液含有0.8微克。培养终止后,加入三倍量的蒸馏水,15分钟后,将细胞低速离心沉淀,用1∶2冰醋酸甲醇固定
This article reports the results of the observation of metaphase chromosomes in 6 patients with malignant reticular cystic disease using bone marrow cells and short-term culture of blood cells. The cell culture method is basically modified according to the short-term culture method reported in Section Dimension et al. The culture medium was treated with Earle’s solution containing 0.5% hydrolyzed milk protein, and AB type human or bovine serum and phytohemagglutinin (PHA) were added. The incubation time was 72 hours. Four hours prior to the termination of culture, colchicine was added so that the culture medium contained 0.8 μg per ml. After the incubation was terminated, three times the amount of distilled water was added and after 15 minutes the cells were pelleted by low speed centrifugation and fixed with 1: 2 glacial acetic acid in methanol
