目的:探讨micro RNA-154(mi R-154)对大鼠肝星状细胞(HSC-T6)凋亡的影响。方法:用合成的mi R-154模拟物(mi R-154 mimics)和mi R-154抑制物(mi R-154 inhibitor)转染HSC-T6,以其阴性对照物(mimics NC、inhibitor NC)转染细胞作为阴性对照。采用CCK-8试剂检测mi R-154对HSC-T6增殖的影响;采用流式细胞术检测mi R-154对HSC-T6细胞周期和凋亡的影响。结果:mi R-154可以显著提高细胞增殖能力,降低细胞凋亡率(P<0.01);细胞内过表达mi R-154可使G0/G1期细胞比例显著下降,S期细胞比例显著升高(P<0.01);抑制细胞内mi R-154表达可显著提高细胞凋亡率,G0/G1期细胞比例显著升高,S期细胞比例显著下降(P<0.01)。结论:micro RNA-154可以促进肝星状细胞的增殖,抑制肝星状细胞的凋亡。
Objective: To investigate the effect of micro RNA-154 (mi R-154) on the apoptosis of rat hepatic stellate cells (HSC-T6). METHODS: HSC-T6 was transfected with synthetic mi R-154 mimics and mi R-154 inhibitor, and its negative control (mimics NC, inhibitor NC) Transfected cells served as negative controls. The effect of mi R-154 on the proliferation of HSC-T6 cells was detected by CCK-8 reagent. The effect of mi R-154 on the cell cycle and apoptosis of HSC-T6 cells was detected by flow cytometry. Results: mi R-154 significantly increased the cell proliferation and decreased the apoptosis rate (P <0.01). Overexpression of mi R-154 significantly decreased the percentage of cells in G0 / G1 phase and significantly increased the percentage of cells in S phase (P <0.01). Inhibition of mi R-154 expression in cells significantly increased the rate of apoptosis, the proportion of cells in G0 / G1 phase increased significantly, and the proportion of cells in S phase decreased significantly (P <0.01). Conclusion: micro RNA-154 can promote the proliferation of hepatic stellate cells and inhibit the apoptosis of hepatic stellate cells.
