目的:探讨透析法制备低相对分子质量人参糖肽的工艺,并对其理化性质、纯度与相对分子质量分布进行研究。方法:以理化性质及收率为考察指标,单因素试验对透析制备低相对分子质量人参糖肽的工艺进行考察,采用HPLC及GPC软件测定纯度与分子量分布。结果:制备工艺确定为采用截流相对分子质量1 000 Da透析袋,加入6 L水透析4 h。制备的人参糖肽总糖肽质量分数92.49%,收率18.95%,确定为相对分子质量在300-3 500 Da的混合物。结论:采用透析法制备低相对分子质量人参糖肽操作简便易行,总糖肽含量较高,相对分子质量较低。
OBJECTIVE: To investigate the preparation of low molecular weight ginseng glycopeptides by dialysis, and to study its physical and chemical properties, purity and relative molecular mass distribution. Methods: The physical and chemical properties and yield as the index, the single factor test dialysis preparation of low molecular weight ginseng glycopeptide process were investigated using HPLC and GPC software to determine the purity and molecular weight distribution. Results: The preparation process was determined to use a cut-off molecular weight of 1 000 Da dialysis bag, dialyzed with 6 L of water for 4 h. The prepared ginseng glycopeptide total glycopeptide mass fraction 92.49%, yield 18.95%, identified as the relative molecular mass of 300-3 500 Da mixture. CONCLUSION: The preparation of low molecular weight ginseng glycopeptide by dialysis is simple and easy to operate with high total glycopeptide content and relatively low molecular weight.
