[目的]研究手性药物氨甲喋呤(MTX)单一对映体对人肝癌细胞株HepG-2的体外增殖的抑制作用。[方法]应用MTT比色法检测MTX对映体[(+)MTX、(-)MTX]对HepG-2细胞增殖抑制作用,用倒置显微镜观察细胞的形态学变化。[结果]0.1~150μmol/L(+)MTX和10~150μmol/L(-)MTX可抑制HepG-2细胞的生长,且其作用具有剂量和时间依赖性(P<0.01),但抑制强度为(+)MTX>(-)MTX;倒置显微镜观察不同浓度(+)MTX和(-)MTX作用HepG-2细胞株不同时间后,细胞出现不同程度的形态学改变。[结论](+)MTX和(-)MTX对HepG-2细胞的抑制作用不同,(+)MTX抗增殖作用明显强于(-)MTX。
[Objective] To study the inhibitory effect of single enantiomer of methotrexate (MTX) on the proliferation of human hepatocellular carcinoma cell line HepG-2 in vitro. [Method] MTT colorimetric assay was used to detect the inhibitory effect of MTX enantiomers [(+) MTX, (-) MTX] on HepG-2 cell proliferation. Morphological changes of HepG-2 cells were observed by inverted microscope. [Results] MTT and MTX inhibited the growth of HepG-2 cells in a dose-and time-dependent manner (P <0.01), but the inhibitory strength of the cells treated with 0.1-150 μmol / L MTX and 10-150 μmol / L MTX were (+) MTX> (-) MTX. The morphological changes of HepG-2 cells were observed under inverted microscope at different concentrations of MTX and (-) MTX for different time. [Conclusion] (+) MTX and (-) MTX have different inhibitory effects on HepG-2 cells. (+) MTX antiproliferative effect is stronger than that of (-) MTX.
