目的研究共刺激分子CD28/B7家族(CD28、CTLA-4、CD80、CD86)mRNA在系统性红斑狼疮(SLE)患者外周血中的表达,探讨其在SLE发病机制中的作用。方法选择44例SLE患者作为病例组、101例健康体检人群作为健康对照组,提取外周血单个核细胞,采用Taqman探针实时荧光定量PCR方法检测CD28、CTLA-4、CD80、CD86 mRNA的表达。结果与健康对照组相比,病例组CD28、CD80、CD86表达均明显下降(P<0.01),CTLA-4表达均有所下调,但差异无统计学意义(P>0.05)。结论 CD28/B7家族共刺激分子的表达降低可能导致SLE患者免疫功能降低。
Objective To study the expression of costimulatory molecules CD28 / B7 (CD28, CTLA-4, CD80, CD86) in the peripheral blood of patients with systemic lupus erythematosus (SLE) and to explore its role in the pathogenesis of SLE. Methods Totally 44 cases of SLE patients were selected as the case group and 101 healthy people as healthy control group. Peripheral blood mononuclear cells were collected. The expression of CD28, CTLA-4, CD80 and CD86 mRNA was detected by Taqman real-time PCR. Results Compared with healthy control group, the expression of CD28, CD80 and CD86 in the case group were significantly decreased (P <0.01), while the expression of CTLA-4 was down-regulated, but the difference was not statistically significant (P> 0.05). Conclusion The decreased expression of CD28 / B7 costimulatory molecules may result in decreased immune function in SLE patients.
