树突状细胞(dendritic cells,DC)作为抗原提呈细胞,具有独特的活化初始型T细胞的能力.白细胞介素-12(IL-12)可促进NK,T细胞的增殖及杀伤活性,以及初始型T细胞向CD4 Thl细胞的分化.本研究采用GM-CSF+TNF-α诱导小鼠骨髓细胞产生的DC,观察了在DC成熟过程中IL-12的产生,以及在DC成熟过程中应用IL-12对DC的表型和功能的影响.采用FLISA法检测了GM-CSF+TNF-α诱生的DC产生IL-12的水平,结果显示,第5天时,10~6DC培养上清中IL-12 P40的水平为120pg/ml,IL-12P70为20pg/ml;至第8天时,lL-12P40和IL-12 P70水平均升高,分别为150pg/ml和24pg/ml.在DC培养的第3~7天,加入IL-12(25ng/ml),可观察到DC在混合白细胞反应过程中,刺激T细胞增殖的能力显著增强;检测DC的表型,可观察到应用IL-12后,CD80的表达水平显著提高,而CD40,CD54,CD80,MHC-Ⅱ等表面分子则无明显变化.上述
