目的:观察脾胃湿热证大鼠胃黏膜细胞凋亡指数(AI)及凋亡调控基因蛋白Bcl-2、P53变化规律,探讨王氏连朴饮治疗脾胃湿热证的作用机制。方法:采用TUNEL法检测胃黏膜组织AI及SABC法同步检测胃黏膜上皮细胞凋亡基因相关蛋白Bcl-2、P53。结果:模型组胃黏膜细胞促凋亡基因相关蛋白过度表达,明显高于正常组及王氏连朴饮各剂量组(P<0.05),正常组与王氏连朴饮各剂量组之间无显著性差异。结论:脾胃湿热证模型组大鼠AI升高,Bcl-2、P53表达增强。王氏连朴饮有抑制胃黏膜细胞凋亡调控基因蛋白Bcl-2及P53过度表达、修复胃黏膜的作用,从而调节细胞凋亡趋于正常。
Objective: To observe the changes of gastric mucosal apoptosis index (AI) and apoptosis-regulating gene Bcl-2, P53 in rats with spleen-stomach damp-heat syndrome and to explore the mechanism of action of treating the spleen-stomach damp-heat syndrome with Methods: The gastric mucosal epithelial cell apoptosis gene Bcl-2 and P53 were simultaneously detected by TUNEL method and SABC method. Results: The overexpression of pro-apoptotic gene-related protein in gastric mucosal cells of model group was significantly higher than that of normal group and Wang Lian Pu Yin (P <0.05). There was no significant difference between the normal group and Wang Lian Pu Yin Significant difference. Conclusion: AI in spleen-stomach damp-heat syndrome model rats increased, and the expression of Bcl-2 and P53 increased. Wang Lian Pu drink can inhibit the gastric mucosa apoptosis regulating gene protein Bcl-2 and P53 overexpression, repair the role of gastric mucosa, and thus regulate apoptosis normal.
