采用荧光和紫外-可见吸收光谱,研究了染料木素与牛血清白蛋白(BSA)的相互作用。结果表明染料木素对BSA有较强的荧光猝灭作用;根据Stern-Volmer方程得到染料木素与BSA之间的结合常数KA为4.37×106(27℃)、6.45×106(37℃)和6.76×106(47℃)。根据F rster非辐射能量转移理论,求出了染料木素与BSA之间的结合距离为2.64 nm(27℃)、2.68 nm(37℃)和2.71 nm(47℃)。热力学数据表明该药物与牛血清白蛋白的相互作用是一个吉布斯自由能降低的自发过程,且二者之间的主要作用力类型为静电引力,同时用同步荧光光谱探讨了染料木素对BSA构象的影响。
The interaction of genistein with bovine serum albumin (BSA) was studied by fluorescence and UV-Vis absorption spectra. The results showed that genistein had a strong fluorescence quenching effect on BSA. According to the Stern-Volmer equation, the binding constants KA between genistein and BSA were 4.37 × 106 (27 ℃), 6.45 × 106 (37 ℃) and 6.76 × 106 (47 ° C). The binding distances between genistein and BSA were found to be 2.64 nm (27 ° C), 2.68 nm (37 ° C) and 2.71 nm (47 ° C), respectively, based on the Frster non-radiative energy transfer theory. Thermodynamic data show that the interaction between the drug and bovine serum albumin is a Gibbs free energy reduced spontaneous process, and the main force between the two types of electrostatic attraction, and simultaneous fluorescence spectroscopy to explore the genistein pairs BSA conformation.
