Intermediate fibrin nestin in a variety of embryonic progenitor cells and mature tissue are expressed in recent years, some studies have shown that the expression of nestin and some pathological features of malignant tumors are related. However, the nestin in stem cells Differentiation and tumorigenesis is unknown. In this study, we used the short hairpin RNA as a tool to study the function of nestin using rat glioma cell line C6 as a model. The results of both trace and migration experiments showed that the nestin gene silencing can promote the migration of C6 cells. At the same time, BrdU infiltration experiments showed that this process was accompanied by the increase of cell proliferation.Further studies showed that the activity of cyclin-dependent kinase cdk5 In addition, the migration induced by nestin gene silencing could be restored by cdk5 specific inhibitor roscovitine, but had no significant effect on cell proliferation. In summary, the present study revealed that nestin Gene silencing is associated with glioma cell migration and proliferation, and cdk5 is an important regulator of this process.
