[目的]建立的刺山柑组织快繁无菌系.[方法]根据不同部位、不同采集时间等,确定刺山柑外植体的采集时间、部位、灭菌剂和灭菌时间.[结果]建立刺山柑的无菌体系时,选择晴天去采集外植体为好;带芽的茎段污染率高,腋芽的污染率低,选用腋芽作为外植体;操作台外的最佳消毒方法是饱和的洗洁精清洗后用自来水冲洗20 min;操作台内消毒对比10%次氯酸钠、2%双氧水、2%溴水、0.1%升汞,0.1%升汞灭菌效果好,而最佳操作台内最佳灭菌体系为75%酒精消毒10 s,以灭菌去离子水冲洗2次,后于0.1%升汞中浸泡9 min,以灭菌去离子水冲洗5次.[结论]对外植体的采集时间晴天相对雨天污染率低,腋芽出芽率高,酒精和升汞双灭菌效果最佳.
