本研究以478玉米自交系及其转基因后代为材料,优化了除草剂筛选转基因玉米后代的方法。结果表明,当除草剂氯磺隆浓度为60mg/L时,利用喷洒的方法可以在田间高效地淘汰大量非转基因植株和ALS(氯磺隆标记)基因表达量低的株系或个体。ALS基因作为除草剂筛选标记基因具有较高的生物活性、选择性强等优点。此方法可以用于转基因玉米后代的辅助筛选。提高转基因玉米后代阳性植株的筛选效率,加快转基因玉米纯合速度和回交转育速度。
In this study, 478 maize inbred lines and their transgenic offspring were used as materials to optimize the method of screening herbicide offspring of transgenic maize. The results showed that when the concentration of herbicide chlorsulfuron was 60 mg / L, a large number of non-transgenic plants and plants or individuals with low expression of ALS (chlorsulfuron) gene could be efficiently eliminated in the field by spraying. ALS gene as a herbicide screening marker gene has high biological activity, selectivity and so on. This method can be used as an adjunct to screening of progeny of transgenic corn. Improve the screening efficiency of the positive plants of transgenic maize progenies, speed up the homozygous speed of the transgenic maize and the speed of backcrossing and transplanting.
