目的了解云南省麻疹野病毒不同基因型的流行分布状况。方法用B95a细胞[Epstein-Barr(EB) Virus-Transformed,Marmoset B Lymphoblastoid Cell Line;埃泼斯坦-巴尔病毒转化的绒猴淋巴母细胞]和Vero/SLAM细胞[Vero Cell Transfected to Express the Human Signaling Lymphocyte Activation Molecule(SLAM),淋巴信号激活因子转染的非洲绿猴肾细胞]从麻疹爆发和散发患者的标本中分离麻疹野病毒,用逆转录-聚合酶链反应扩增核蛋白(Nucleoprotein,N)基因羧基末端的676个核苷酸片段,对扩增产物进行核苷酸序列测定。并以N基因羧基末端450个核苷酸序列构建基因亲缘性关系树,进行核苷酸变异分析。结果云南省2004~2011年共分离和检测到46株麻疹病毒及2株病毒核酸,43株为H1基因型,分布于14个县(区)及四川省普格县;4株为d11基因型,分布于边境孟连县及缅甸的班康市;1株为A基因型,来自龙陵县。结论麻疹病毒H1基因型是云南省优势本土基因型,d11基因型是从缅甸输入后连续两年于缅甸接壤的地区发现的麻疹野病毒,A基因型为中国沪191疫苗株相似株。
Objective To understand the epidemiological distribution of different genotypes of wild measles virus in Yunnan Province. Methods Vero Cell Transfected to Express the Human Signaling Lymphocyte (Vero Cell Transfected to Express the Human Membrane Lymphocyte Cell Line; Ehrlich-Barr Virus-transformed Marrow Lymphoblastoid Cells) and Vero / SLAM Cells [Epstein-Barr Activation Molecule (SLAM), a green fluorescent mouse kidney cell transfected with lymphotoxin, isolated wild measles virus from specimens of patients with measles eruption and dissemination, and amplified nucleoprotein (N) by reverse transcription polymerase chain reaction (RT-PCR) The 676-nucleotide fragment of the carboxyl terminus of the gene was used to determine the nucleotide sequence of the amplified product. Nucleotide sequence analysis of 450 nucleotide sequences of the N gene was used to construct the phylogenetic tree. Results 46 strains of measles virus and 2 strains of virus nucleic acid were isolated and detected in Yunnan province from 2004 to 2011. 43 strains were H1 genotype and distributed in 14 counties (county) and Puge county in Sichuan Province. 4 strains were d11 genotype , Distributed in the border Menglian County and Burkina Faso City; a strain of A genotype, from Longling County. Conclusion The H1 genotype of measles virus is the predominant indigenous genotype in Yunnan Province. The d11 genotype is the wild measles virus found in the area bordering Myanmar for two consecutive years after importation from Myanmar. The genotype A is the similar strain of Shanghai-191 vaccine strain in China.
