MicroRNAs(miRNAs)是一类非编码小分子RNA,是体内重要的内源性基因调节因子,主要是在转录后水平调控靶基因的表达。本研究是通过人工microRNA技术(artificial microRNA,简称amiRNA),与PCR技术克隆得到了701 bp的miR159片段,然后将获得的miR159片段通过双酶切后链接到植物表达载体p CPB121,构建了马铃薯amiR159表达载体。经PCR和酶切分析鉴定,确认载体构建成功。为后期通过马铃薯的遗传转化研究miR159/MYB的调控模式提供了帮助。
MicroRNAs (miRNAs) are a class of non-coding small RNAs that are important endogenous gene regulators in the body and mainly regulate the expression of target genes at post-transcriptional level. In this study, a 701 bp fragment of miR159 was cloned by artificial microRNA (abbreviated as amiRNA) and cloned by PCR. The resulting fragment of miR159 was double-digested and ligated into the plant expression vector pCPB121 to construct the amiR159 Expression vector. PCR and restriction analysis confirmed that the vector was constructed successfully. This provided help for studying the regulation pattern of miR159 / MYB through the genetic transformation of potato.
