目的 研究正常中国男性雄激素受体 (androgen receptor,AR)基因 (CAG) n重复序列的多态性。方法 应用 DNA测序基础上的 [α- 3 2 P]d CTP掺入不对称 PCR-变性聚丙烯酰胺凝胶电泳(polyacrylamide gel electrophoresis,PAGE)方法 ,对 10 7名正常男性 AR基因的 (CAG) n重复数进行测定。结果 AR基因 (CAG) n序列在正常男性人群中呈现重复多态性 ,其重复范围为 11~ 2 9,集中于 2 0~2 4,以 2 2最多。结论 AR基因 (CAG) n序列在正常男性人群中呈现重复多态性 ,为今后进一步研究其病理学及遗传学意义打下基础。
Objective To study the polymorphism of the repeat sequence of androgen receptor (AR) gene (CAG) in normal Chinese male. Methods A total of 10 normal male AR genes (CAGs) were obtained by asymmetric PCR-polyacrylamide gel electrophoresis (PAGE) with [α- 3 2 P] d CTP incorporation by DNA sequencing. n repeat determination. Results The sequence of AR gene (CAG) n showed polymorphism in normal male population. The repeat range was 11-2 9, with a concentration of 20-24, with a maximum of 22. Conclusion AR gene (CAG) n sequence shows polymorphism in normal male population, which lays the foundation for further study of its pathological and genetic significance in the future.
