目的:研究在Sertoli细胞分化和成熟阶段,外源性雌激素对Sertoli细胞FasL表达以及血睾屏障的影响,进一步讨论影响血睾屏障的相关因素及其可能机制。方法:体内体外分别给与青春期SD大鼠大剂量的雌激素(二乙烯雌酚和雌二醇),运用免疫荧光方法和Western印迹观察Sertoli细胞内FasL的表达以及电镜下血睾屏障结构的变化。结果:外源性大剂量雌激素作用下,Sertoli细胞内FasL表达量明显增强(P<0.05);同时出现Sertoli细胞膜和生精上皮血睾屏障部位FasL的表达量上升;在18d实验组,电镜显示示踪剂镧透过血睾屏障,到达生精上皮全层。结论:外源性大剂量雌激素能使Sertoli细胞内FasL的表达量和分布区域发生改变并影响血睾屏障的结构。
OBJECTIVE: To study the effect of exogenous estrogen on FasL expression and blood-testis barrier in Sertoli cells during the differentiation and maturation stages of Sertoli cells, and to further discuss the related factors and their possible mechanisms. Methods: The estrogen (diethylstilbestrol and estradiol) were given to adolescent SD rats in vitro and in vivo respectively. The expression of FasL in Sertoli cells and the changes of the structure of blood-testis barrier under electron microscope were observed by immunofluorescence and Western blot. . Results: The expression of FasL in Sertoli cells was significantly increased under the exogenous high-dose estrogen (P <0.05). At the same time, the expression of FasL in Sertoli cell membrane and the blood-testis barrier of seminiferous epithelium increased. In the 18th day, The lanthanum tracer is shown to penetrate the blood-testis barrier and reach the full layer of the seminiferous epithelium. Conclusion: Exogenous high-dose estrogen can change the expression and distribution of FasL in Sertoli cells and affect the structure of blood-testis barrier.
