目的 观察不同浓度As2 O3 对结肠癌Lovo、LS 174T细胞株生长的影响。方法 0 12 5~ 2 μmo1/L的As2 O3 与结肠癌细胞株Lovo、LS 174T共同孵育 ,观察不同浓度As2 O3 作用不同时间对结肠癌细胞的生长抑制作用、细胞形态学改变、DNA的变化。结果 1~ 2 μmol/LAs2 O3 作用的结肠癌细胞呈凋亡特征性改变 :细胞膜不发生破裂 ,细胞核出现核浓缩、核碎裂和凋亡小体形成 ;流式细胞仪检测在G1期前出现亚二倍体凋亡峰 ;细胞DNA抽提琼脂糖凝胶电泳呈凋亡特征性条带。结论 1~ 2 μmol/LAs2 O3 可诱导结肠癌细胞凋亡 ,肿瘤细胞凋亡与Fas、Fas L表达有关
Objective To observe the effects of different concentrations of As2O3 on colon cancer Lovo, LS 174T cell growth. Methods The As2O3 of 0 12 5~2 μmol1/L was incubated with colon cancer cell lines Lovo and LS 174T. The growth inhibition, morphological changes and DNA changes of colon cancer cells at different concentrations of As2O3 were observed. RESULTS: Colon cancer cells treated with 1-2 μmol/L ars2 O3 showed characteristic changes in apoptosis: cell membranes did not rupture, nuclei appeared nuclear condensation, nuclear fragmentation, and apoptotic bodies; flow cytometry was detected before the G1 phase. Subdiploid apoptotic peak; cell DNA extraction agarose gel electrophoresis showed apoptotic characteristic bands. Conclusion Apoptosis of colon cancer cells can be induced by 1~2 μmol/LAs2 O3. The apoptosis of tumor cells is related to the expression of Fas and Fas L.
