目的探讨开郁清胃颗粒改善OLETF大鼠模型胰岛素抵抗作用的机制。方法32周龄雄性OLETF实验大鼠随机分成4组:模型组、罗格列酮组、开郁清胃颗粒大、小剂量组。正常对照组选用同周龄Wistar大鼠。各组分别给药4周行正常血糖高胰岛素钳夹实验后检测空腹血糖、胰岛素、胰高血糖素、皮质醇、胃动素、胰岛素样生长因子-1(IGF-1)、甘油三酯和胆固醇含量的变化,胰腺切片HE染色观察组织学改变。结果开郁清胃颗粒对OLETF大鼠空腹血糖、胰岛素、胰高血糖素和胃动素无明显影响,但能降低OLETF大鼠的皮质醇、甘油三酯和胆固醇水平;升高胰岛素样生长因子-1水平。OLETF模型组胰岛表现为脂肪变性、炎细胞浸润和纤维增生,开郁清胃颗粒大、小剂量组胰岛结构明显好于模型组。结论开郁清胃颗粒能降低OLETF大鼠的胰岛素抵抗的作用机理与升高IGF-1,降低皮质醇、甘油三酯和胆固醇水平有关,并能有效地保护OLETF鼠胰腺组织形态结构。
Objective To investigate the mechanism of Kaiyuqingwei granule in improving insulin resistance in OLETF rat model. Methods Male OLETF rats at 32 weeks of age were randomly divided into 4 groups: model group, rosiglitazone group and Kaiyuqing stomach granule large and small dose groups. Normal control group Wistar rats of the same age were used. The rats in each group were administered with normal blood glucose and high insulin for 4 weeks. Fasting blood glucose, insulin, glucagon, cortisol, motilin, insulin-like growth factor-1 (IGF- Cholesterol content changes, histological changes were observed by HE staining of pancreatic sections. Results Kaiyuqingwei granule had no significant effect on fasting blood glucose, insulin, glucagon and motilin in OLETF rats, but could reduce the levels of cortisol, triglyceride and cholesterol in OLETF rats, and increase insulin-like growth factor -1 level. The islets of OLETF model group showed steatosis, infiltration of inflammatory cells and fibrosis. The islet structure of Kaiyuqing stomach granule in large and small dose groups was significantly better than that in model group. Conclusion The mechanism of Kaiyuqingwei Granule in reducing insulin resistance in OLETF rats is related to increasing IGF-1, decreasing cortisol, triglyceride and cholesterol levels, and effectively protecting the morphology of pancreatic tissues in OLETF rats.
